A cell's reduction in the number of a given receptor in response to prolonged exposure to a substance that activates it — a normal adaptive process, not unique to psychedelics.

Example: Daily SSRI use down-regulates 5-HT2A receptors over time, which is the leading explanation for why people on therapeutic-dose SSRIs often report feeling little from a standard psilocybin dose — see Drug Interaction below.
